Default Coolant air bleed in rear radiator pickup?

I'm putting an ls1 into a toyota prerunner pickup and plan to place the radiator behind the cab (no glass in cab) with the main coolant lines running under the cab. I'm unsure as to how I should treat the coolant air bleed that usually runs a small hose back to the top of the radiator. Can I simply bleed out all the air once the system is full and then plug it off or will air pockets develop?
Advice needed thanks.

Needs to be vented to a low pressure area of the closed cooling loop. Top radiator hose or some people drill a hole in the top of the water pump any convenient place, albeit, it really should be a higher point then the head pick off spot.
