What goes in the temp sensor sized hole on the passenger side head? Is a plug supposed to be in there?

yes you can plug it or run a redundant temp sensor for an AutoMeter temp gauge or any brand. I like it just for the backup.

That is a drain plug for flushing the cooling system. You take it out and run water into the system until it comes out clear.
