I have a 2000 camaro ss and I just got Hooker Lts and a Mufflex y-pipe and I was wondering should I get it tuned.

Yes I would get it tuned or you'll be wasting your money on your headers.

Yes, you should get it tuned. The AFR will be off a tad and are you using cats? if not, have the COT disabled along with the codes.

Your Air fule may not be off at all, some stock cars run a little rich and with the headers you may be righ in the 12's. But I would deff. consider it you can add more timming and remove the tourque managment, or adjust if it's a auto, and set the airfule to a nice 12.6.

Can I tune my car with a hypertech or do I need to let a f-body specialist tune it.

Hypertech will not do what you would want. It is best to let a tuner do it, or best yet get yourself one of the better softwares (like HPTuners ) to tune it. Cost will be comparable to having it tuned by a shop.
