Manual focus tips.. A couple camera Q's too..

No matter how hard i try I still seem to only get a few good shots w/ the manual focus. Do I just focus on the subject??

What should I have the foolwing set to??
Contrast, Sharpness, Saturation, and Noise Reduction.. I have all set @ standard, xcept sharpness.. That's on High...
