Most shops advise 250-500 miles, but I've never bothered to break one in. Drive it for one or two miles, let the fluid circulate, make sure it shifts OK, double check the fluid, and let 'er rip.

After spending a month in my garage building the 4L80-E the first day it was in the truck I was doing burnout and WOT sprints to tripple-digit speeds. It's still as perfect as it was that day a year ago. Of course I did make sure it was shifting right and the fluid was dead-on perfect. The 80s are not as sensitive to absolute fluid level as the 60s are, better to be safe though.
