Blow-Off Valve = Purges excess air pressure from the cold side of a boosted application if pressure levels are exceeded. Applications: Any system that increase manifold pressure above atmospheric.

Wastegate = Purges excessive exhaust pressure prior to the turbine how to regulate required boost. Applications: Any exhaust driven aircharger (Turbos).

You can run without both, but you run the risk of damaging the compressor blades without a blow-off valve, or having excessive boost pressure without a wastegate.

Unless you get a ridiculous AR turbine housing that never really allows you to spool up, you will have to have a wastegate. The Buick GN's run stock without BOV's, but it is hard on the thrust bearing and will decrease the life span of the turbo. After the thrust bearing goes, you will destroy the compressor or turbine (essentially time for a new turbo at that point)
