when I install a stroker kit let say 402 on a ls2 block (aluminum)
Do I need to worry about the block it self

Not unless your putting 15+psi of boost or a 300 shot of sauce on it.

Are you asking if you need to machine and clearance the block when putting the rotating assembly in? The answer to that is a definite YES!
