Can I run a 2010 4L60e with a Gen III engine?
I assume it will bolt up ok, so it may boil down to: Can/will a Gen III ECM control a Gen IV vintage tranny??

Short answer - No.
In fact a 2010 4L60E will require a 2008 or later PCM (e.g. TCM) due to significant changes that were made in 2008, including deleting the 3-2 Shift solenoid and replacing the pressure switch (on the valve body) with a mechanical switch on the internal shift linkage.
Several members here have tried to make it work by deleting codes and stuff, but without success.
I am running a 2010 4l65E with a 1998 PCM, but this required replacing the valve body with a valve body from a 1998-2006 trans. I am helping (via email and phone) another member with that conversion now and will he happy to help others too. You will need a good 1998-2006 valve body (preferably rebuilt), the internal harness and the pressure switch. These are always available on ebay for around $175. You also need a Transgo separator plate with gaskets for another $30.
Having the valve body removed is a good time to install a few other parts or even the Transgo HD2 shift kit; the HD2 is designed for '96 to '06 transmissions, but with one mod it will work in your 2010 too. The mod is: DO NOT install the "alum spacer" on the boost valve or your line pressure will be too high. There are more details than I care to cover here unless there is interest in them.

The changes in 2008 affected the 4L60E, 4L65E and the new 4L70E. I didn't see any obvious improvements except for perhaps better sprags.
The purpose for the 3-2 shift (PWM) solenoid was to soften this downshift. Since all 2008+ cars have drive-by-wire, the ECM/TCM could now soften the downshift by modulating the throttle and those transmission parts were no longer needed. I have no idea why they changed from a pressure switch to a mechanical switch.
Note that the hardened sun shell was introduced somewhere between 2002 and 2003. Other improvements were made around that time too.
