Default 4L80E clunk

Little info.
The motor and trans are brand new. I am in the process of tuning the engine. The trans is a built unit. The line pressure while in gear applying throttle sets between 195-200.

I want to put vacuum on the motor to help seat the rings. I was driving the car down the road and when I shift from Drive to Overdrive I get a clunk sound under light throttle. Going from OD to D. Everything is fine. Going from D to 2nd everything is fine. It sounds like it is coming from the transmission. What could this noise be? The bell housing bolts are tight, the converter bolts are tight, the cross member and bolts are tight. I am running a poly mount. Trying to get an idea before I call the trans builder.

Thanks

No way its a joint. I just had brand new solid joints installed. I will disable the convert and take it for a drive tonight.
