if the weight is greater it will affect as there is more rotational mass.
getting real technical, the bigger the rim, even if its the same weight as a, say, stock 16" wheel will create *some* loss as the rotational mass is further from the wheel center. i would imagine it would be very little however.
now dynoing with a slick will result in a loss too... even a lightweight rim/tire setup...
