If I'm running new rails and an aeromotive regulator up front with a return line, can I use the vapor line as a return line back into the gas tank? If not, how do people generally return the fuel back to the tank?

If the vapor line is big enough in diameter... But thats if its safe for raw fuel to be in that vapor line.
Also, you'd then need to somehow vent the tank with another hose and check valve.

I used a -6an braided line for my return... And since all my pump stuff was removed from the sending unit, I clamped the -6an line right to the old 'outlet' tube. Which just required me to take that plastic retainer clip off the hard line so my hose could go on far enough.
