Poly motor mounts= yes. I have them and love them. If you have long tubes they are a must!
Trans mount= bad. I've heard they can crack the tranny case. I would just keep the stock rubber tranny mount.

What he said. Before I got the poly motor mounts, my pacesetter ory would beat the hell out of my floorboard. After the mounts, not one bang from the y pipe!! Plus it helps get the power to the rear wheels. But they are a huge pita to install.

Some header designs require it as they are a garbage design. I have Dynatechs with their Catted Y and never an issue with Stock Rubber on both the engine and trans. I did put a TQ arm relocation kit on the car and just did the Poly on the trans mount. Now I got a decent vibration from 70ish and up and now I gotta track that down and fix it. I have been told its due to the preload plate. Who knows.
With a stiffer mount, vibrations do transfer but will keep flexing down.
