I have AI ported 226cc 243 heads with stock chambers, roughly 65cc's. The heads have a BTR spring kit in them.

My cam is a custom grind from Tick. 231/235 .636in .612ex 112+4

I plan on using cometic .040 head gaskets instead of the stock .051 for a little higher compression.

Obviously measurements will be necessary, but should my PTV clearance be ok without flycutting for this cam? The bottom end is completely stock.

You should be completely fine, but it's always the right move to measure. Cams like the Polluter, T-Rex, Vindicator, Hellion, etc. are all designed to clear stock, unmilled heads, so you should be more than okay.

Thanks....my tuner questioned me about it. He said I might run into some clearance issues, but when Martin spec'ed it out he knew what my plan was. He told me I could run that head gasket, but then when someone else I respect questioned it I started second guessing.

If the heads were milled then I could understand my tuner's concern.

Your tuner is just trying to be safe, which I don't blame him. If that **** goes "kaboom", it's going to likely fall back on him, when in all actuality, it would likely have nothing to do with what he did at all. I'm sure your tuner would feel much better if you could give him the intake and exhaust clearance numbers.
