What are the physical differences in the geniii blocks and the geniv blocks

On any given sized block the big differences are the knock sensor and cam sensor locations. The gen 3 runs the knock sensors off the valley cover, and the gen 4 runs them off the side of the block. The cam sensor on the gen 3 runs off the back of the motor and the gen 4 has it on the timing cover

can the gen 3 sensors be used in the gen 4 block?

You have to bore the holes out in the side of the block(the ls1/gen 3 style sensors have a larger thread pitch) and then get the correct harness. Racetronix has a harness i believe.

Gen IV engines also have Active Fuel Management capabilities.
