A 180 stat with a 160 tune will just make the fans run a lot, pretty much continuous. The loss of coolant is a seperate issue.

Being "tuned for a 160 tstat" just means that the temperature at which the fans turn on are adjusted so that they work well with the thermostat. For example, being "tuned for a 160 tstat" vs being "tuned for a 180 tstat" just would mean that the cooling fans on the radiator would come on approx 20 degrees cooler or so. (this is just an example, I'm not familiar with LT1 tunes, just other cars)

This would have no direct affect on coolant. Fill the radiator back up to the top with the proper matching fluid. Make sure the overflow is at its proper level as well. If you drive it around for awhile and the level drops, you have a leak somewhere.
