I will be installing longtube headers, 3 inch ORY with slp loudmouth next week. I was wondering what needs to be changed in the tune once its installed? Do I just have to change o2 sensor 2 dtcs from coming on or is there more to it?

The rear O2's will need to be deleted. That is all you should really NEED to do but to get the most out of it you'll want to play with the MAF, VE and timing tables. If it's a totally stock tune you'd probably gain 20hp from a tune after the exhaust.
