You can keep the ECM and it's programming that's in the truck already as it's programmed for the truck and all of it's options. The 6 speed tranny will shift and operate normally.
To get the TUTD working it is a lot more involved because in those years only the uplevel trim trucks got the 10 speed. Since you have a donor you are already time and money ahead.
To get the TUTD working you need to swap these module from the donor ECM, BCM, stock radio if you are stock, xm module if you are using it, and rear object detection module if your truck has it
You can either swap the donor shifter to your truck or swap the complete column
You can straight swap in the ins cluster it's plug and play and you will Have D and M in the display as opposed to 1234 on the 4 speed cluster.
If you move everything over from the donor it wont know the difference when you hit the key it will think it's business as usual. Depending on how your truck is optioned there may be a few things to iron out. It really depends on what trim level your truck is and how it decides to play along.

The reason for swapping the BCM and all the interior components is BCM controls TUTD and broadcasts bus infor to the ECM TCM. The other interior modules are vin locked and will not operate once the BCM is changed without doing GM level programming on everything which can be done. But since you have a whole truck at your disposal swap it all and save yourself some headache. Then your existing truck uses the 09 vin information for all of those modules going forwards in the future. So be dang sure to write that number down somewhere safe and save it for a rainy day in the future when a module dies and has to be reprogrammed
