I have a 2000 WS6 and noticed the car is running a little rich after installing. Will having it tuned for this make a big difference?

Yes, it will make a difference. The PCM is computing injector timing using the stock 26# injectors curves and you now have 28#, so 7% more fuel is going in at a given rpm. I'm sure the PCM will eventually be able to learn this out w/ the LTFTs, but I'd still get it tuned.
