The back side of the y would give you the best reading, but if you want to take the readings again after you would want to take it from the collectors.
BTW, there are tools that mount in the AIR pump or O2 fittings, which might be the easiest approach.

I've never heard of this. How can it work since there's still a flowpath through the motor past the rings or past the exhaust valves that are open and out the throttlebody. I mean the TB will be shut but there's still flow through the hole in it. I would think you need the flowpath up the exhaust towards the motor sealed to make it more accurate.
edit - nevermind, this is probably done with the motor running right?

You basically want to run the engine at wot up to your target RPM. I believe the target is 7PSI or less and for every 1 PSI above that figure about 5hp loss.
