fuel line advice

Hi folks, another newbie here so all help is appreciated.
I am putting a 2006 5.3 liter Vortec in my '53 GMC pickup.
I will be using the truck manifold. My question is what configuration of fuel lines will I need to run? The motor has 2 fuel lines that exit the engine compartment on the driver's side. One is connected to the fuel rails but the other is connected to a fitting [plastic] right behind the Throttle body and follows back to the feed line. One line set up? Two lines?

it should be a non-return style rail (use corvette filter/regulator) and the plastic line is evap

Plug evap line at manifold and remove? Just want to be clear. Thanks
