one of 2 reasons. either pinion bearing is excessively worn and starting to howl. Or in my case last spring your ring or pinion gears teeth are starting to crack. My ring gear had 6 teeth that were cracked when i tore it down. The cracks were at the base of the teeth and were hairline thin. That was on the stock 3.23's in my 00 ws6 a4. Now have 3.73's.

I would try changing the fluid first, and when you take the cover off look at the teeth to see if they are worn. If you see nothing wrong and it still makes noise take it to a shop to check the pinion bearing and the runout.
