PCV helps eliminate harmful vapors inside your engine. It sends them through the intake to be burned off. That hose you are referring to is bad on just about every LS1. They just don't withstand the test of time on these engines. I would suggest patching it up with some new hose, buying an intact used unit, or buying an LS6 valley. I like the 3rd option the best.

I just noticed the same thing on mine. I'm probably going to find an in tact used replacement. I talked to the dealership here and they said the only LS6 intake around is in Louisiana. Did GM stop producing them?

From what little research I've done, it'll be fine to drive, but it has a very real possibility of making quite the mess in your engine bay. I wouldn't do it for long unless you wanna change your oil every 800 miles or so as well, because it apparently has a lot to do with keeping your oil from looking like brownie batter.
I guess what I'm getting at is go ahead and reassemble everything, block the fittings on your intake and TB, and drive it to the parts house to get the parts mentioned for the fix above, or drive it to the dealer for the replacement hose, which IIRC is about $130. I wouldn't plan on driving it like that for a while though.
But I'd probably get some sort of affirmation before doing this, as I am far from an expert on the topic.

Short term it gets a little sloppy under the hood. I would not block the system off completely because your engine needs to Breath. There was an old thig I think they called Vapor lock before the PCV.

Only blocking the hole in the intake. It breathes plenty. The holes in the valve covers are open. I drove mine up to my dad's shop where I'm doing a header install tomorrow and it did fine. I plan to have the PCV system repaired by tuesday, though.
