If I remember correctly running rich will affect emissions, fuel economy, and power in that order so yeah it could affect the acceleration.

I haven't driven an auto in a while but aren't 30 and 65 at the top of 1st and 2nd gears respectively and 45 is at the bottom of 2nd? So when you punch it at 45, it's not going to accelerate very well in the bottom of 2nd in a stock auto...
