Default LS2 short block w/ LS6 heads?

Will an LS2 short block work with LS6 heads, intake, oil pan, timing cover, cam sensor, A/C. P/S. accessories etc.?

Will EVERYTHING bolt up?

I'm not sure what you are saying but the stock factory LS2 has 243 heads that was used on the LS6.

Yes and No.

Yes they have the 243 LS6 head " casting " but not the LS6 " light-weight " valves.

And no, the LS6 is the Gen III. LS2 is Gen IV. So your valley cover, front & rear covers won't match.

No, front and rear, and valley cover might not, but everything you asked about will work. So the answer to you question is.....................................

YES

Default LS1 to LS2 = Easy

I swapped a LS2 into my C5. Relocate the knock sessors, a few harnesses and covers and your done. No big deal.
