I say 441 ci motor...once the lsx intake and AFR heads come out, those extra cubes will really make a difference.
I have been waiting ALL to long for them to start making LS1 heads. Rumor mill is saying that they are making 300cfm @ .600 on thier test heads.
I was hearing rumors of something silly like 350cfm heads as the goal with porting

AFR will be making a new casting correct? And if so 300cfm @.600 lift ot of the box is awsome. do some porting and bang the big numbers

In my opinion any stroke over 4.00 is too much for a LS1/6 motor as i have heard from several reliable sources that some of the 4.125 cranks are causing oil consumption and no of at least one case personally where this was true. Also, i believe the geometry of the lS1 motor is not a desirable if you go bigger than 4.00 crank.
Also, stick with a LUNATI crank b/c i also know personally of a new 4.125 CALLIES crank where the welds got loose and the crank had a reluctor wheel problem similar to what PSJOHN had and in my opinion Callies has not perfected the LS1 motor and LUNATI has. Stick with LUNATI and keep it a 4.00 crank for a 427 motor, as that is a more battled tested proven performer vs. the bigger strokes.
Do what GM CHEVY racing does, 427 cubes with a 4.125 bore and 4.00 crank.

2001CamaroGuy, Mark @ AAP had the crank cryoed somewhere here in SoCal, I don't know where though. I'm sure if you call around there are some places that would do it. I'm also running a 6.125 rod. As far as track time goes I currently have the !money mod Still need to get the 5 point cage welded in and get my 28" tires for the track. They will be coming soon though.

I personally know of a good friend who went with a brand new 4.125 CALLIES crank in a LS6 motor and after only 2,500.00 miles, the welds on the crank broke and the reluctor wheel came loose, causing the engine to cut out in the higher rpms.
I have been told by the most reliable of sources, that the CALLIES cranks for whatever reason that is way above my comprehension is more likely to do this than a LUNATI crank and to stick with a LUNATI crank and go with no more than a 4.00 crank. Hence my suggetion this gentleman go with a proven bore and stroke size that being 4.125 bore and 4.00 crank for 428 Beatiful cubes rounded off to 427 of course!

What about weight and balancing? Are the eagle rods and callies crank heavier than the lunati stuff? When the crank and rods are heavier what effect does that have on the pistons? Do they need to be heavier to balance the engine or do they need to be lighter?
