TDC vs Base circle

When a cylinder is at TDC, does that mean the cam is also at base circle for that cylinder? If not, how do you determine base circle on cam? (For setting lash to zero)

First question: yes and no. Both valves are open at TDC between the exhaust and intake strokes, but they'll both be closed between the compression and power strokes.

Second question: EOIC Method. Google it.
