Just a quick note because I could not easily find such info when I was looking for it.
Last night, using the internet based GM SPS service flash system, I was able to reflash the operating system in my 6L80e trans on the bench from 2007 to 2009, using the Speartech harness and a GM MDI interface. I did have an E67 ECU on the harness as well, but it was a different VIN number (this was my core question). GM charges $40 per VIN number for downloads.

Not yet. I'm following a thread on EFI Live where a guy has though, so same thing. I just "read" the stock code with HP Tuners yesterday while the trans is on the bench, it read fine, and shows me what parameters are editable. Shift points, line pressure, torque converter lock/unlock, etc... Should be fine. Main point few are saying is that the ECM and TCM operating system years really need to be the same, so that is why I upflashed it from a 2007 to 2009, because my E67 is a 2009.
It should work OK, I next need to disassemble the entire trans to get the output shaft out to have it machined down from 32 spline to 27, to re-use my existing Colorado transfer case. I am custom making the 6L80 to Colorado transfer case adapter since nobody makes it.
