A 4L65E is just a 4L60E with upgraded bands/clutches and 5 pinion planetary gears. Maybe one or two other upgrades from the factory. They're the exact same transmission in reality.
A 4L80E weighs I think something like 50-70 lbs. more than a 4L60E. Don't quote me on that. I don't know the exact number. It's more than 50lbs. but less than 100lbs.

The 4L80E will hold gobs more horsepower than the 4L60E. The 4L80E when built right will easily take you north of 1000rwhp. Those transmissions were built for 6,000 lb. busses. A 3500 lb. street car is a piece of cake for them.
That being said, they also weigh a lot more than a 4L60E, eat up a lot more horsepower due to parasitic losses and need modifications to fit, making the swap somewhat difficult and more costly. If you have a 1000 rwhp street car and you need overdrive, they're pretty much your only option, but for the vast majority of guys on this site, they're overkill and will just weigh you down and eat up your power.
In short, what is better depends on your application.
