I think that might be there for a PCV port. Most builds just seem to plug that off.
Make sure you put sealed screws in the holes for mounting the box. They go through and will cause a vac leak if they aren't blocked off.
I have a PCV and brake booster hooked to the back of the carb and the MAP sensor hooked to an unported vac nipple on the front of the carb.

I got the carb info from an old guy at NAPA, he gave me some bad info. I got my old Holley book out and looked it up.
Carb ports
Large Port on back of carb
CV hose/brake vacuum hose
Vacuum port in R/side of carb metering block: Timed vacuum port for vacuum advance
Vacuum port on R/side of throttle plate: Full time vacuum for auto trans
Vacuum on Intake can also be for auto trans
