does anyone know if i can mill my heads .040 with a cam gross lift of .616/.621?

I don't know the answer to your question.

Will a 241 flow well enough to benefit from that much lift? If you're going through the trouble to take the heads off to mill them maybe you should just upgrade to a better head.

lift has nothing to do with PTV (Tappet lift does but that is a different story).

What durations/LSA/ICL

Rarely can you ask the question that is this broad about clearance and not get an answer that says "measure". You will have the heads off, so there is no reason not to manually check ptv.
