You messure with zero lash. If it is a hydro lifter then mesure at zero lash. Once you think you have the hight right, take a sharpy and color the top of the valve stem. Place the pushrod length checker you already set in the lifter and put your rockers on and you tighten everything down. Now rotate the engine so that the valve you are working on opens and closes and couple of times. While it is opening and closing check to make sure the spring on the valve is not coil binding. once you have turned it a couple of times take the rocker off and check the wear on the top of the valve stem and make sure it is in the middle and not on the outside, or inside of the stem. Then buy your push rods and do this all over again once you have the actual pushrods and check the wear pattern on the valve stem. Then reasymble and your done.

By the way you can try this with your current pushrods and see if they are the right length.

I have found that the best stuff to paint the valve tip with is ring and pinion gear marking compound. Works way better than the sharpie. Next best thing is a dry erase marker.

You will need a solid lifter or a very weak valve spring, the pressure of the normal spring will collapse the lifter and you won't get a true reading.
