Personally I would go this route:
Junkyard 5.3 motor = $500+Head studs,gaskets & springs minimum
Brand new S475 turbo from forcedinductions.com = $650Plus oil feed block, return and supply lines
Odds and ends to complete turbo kit and fuel system = $1000I wish, good gate and BOV will run more than 500 to control that S475. Plus flanges, truck manifolds, all piping for hotside, all cold side + intercooler, injectors are easy $300, in tank pump set up, random couplers, fab down pipe and connect to exhaust. OP has a clean WS6 doubt hes going to dump it through the fender any relocation brackets he'll need
Bam, for $2150 you have a 650rwhp capable car. Done and done.

Min 4" bore for LS3's. You could probably sell your cam and ported 243's and about break even on a LS3 head/cam set up for a 6.0. You'll need a LS3 specific cam grind since they like a larger split. also need LS3 rockers and valve covers though.
Your 243's and cam would probably make good power on a LQ9/LS2, LQ4 being alittle less due to compression. LQ9/LQ4 is a drop in set up aside from drilling 1 alternator bolt hole. Any LS2 or Gen 4 set up will require conversion of cam sensor, relocate knock sensors, and pending year 58x to 24x reluctor wheel box, about $300. L92/LS3 will require LS2 front cover etc plus everything else I listed.

I was able to nab a set of L92 heads with rockers and stands for $575. I also nabbed a complete LS3 intake sans TB for $255...... The deals are out there, just gotta look.
I have a 6.0 LQ4 that will be getting this top end. I got the LQ4 shortblock for a spare LS1 bare block i had laying around.
If you can find parts such as your ported 243's and cam to sell/trade you can get everything relatively easily w/o a crazy amount out of pocket. Also yes a custom cam would be a great idea. PatG or PredZ would both be able to help you. I ordered specs for a cam from both

I got it a few years ago. Some dude was putting a LS2 in his camaro and sold me the LS1 block, crank, pistons/rods for $200. A few months after that, I was able to trade the LS1 bare block for the LQ4 shortblock. Then sell the crank $100 and the rods/pistons for $115. So I essentially got it for free.
To buy a LQ4 shortblock you are going to pay ~$450 and for a LQ9 you are looking at ~$700 (from what i've seen around)
Or you can get a LQ4 and LS2 rods/pistons for ~$100-150 and save a bit.
