I assume you are talking about from a roll. In that case, I think it is best to kick it out of OD and into 3rd. I hear it is hard on it to shift out of OD at WOT (and it seems to take a loooong time).
Then what I do (not sure if it's good/bad) depending on my speed/rpm, I may kick it down a gear or two so I don't have to wait for the downshift. It seems that M6s usually get the jump on me while the tranny is downshifting. But as soon as I go WOT, I bump it back into 3rd.

If you have the trans settings tuned right there is no
way you'll beat the PCM, and no way a M6 will match
the shift quickness / consistency. If you have a proper
torque converter the M6 won't beat you off the line
either (unless your tires suck). On the other hand a
stock/untuned A4 has suck-*** shiftpoint settings
and someone who knows the setup, will want you to
go from a roll that puts you in a crappy starting spot.
So lie.
Street racing is bad. The Man is out to get you and
Mr. Oak Tree would like a piece too. Best find a smarter
place to learn the limits of your abilities and your luck.
You'll meet a better crowd at the track, and not lose
your license beating some unworthy ***-clown in a
fart-can ratmobile.
Only ricers hand-shift an electronically controlled
automatic. Because they have the mad skillz, yo.

if im above 30mph and below 40mph
and i stomp the throttle
somtimes it will downshift to 2 instead of 1
so now i manually drop it in 1 using the gear selector
but i put it right back up to 3 right after.
it can handle upshifting perfectly on its own.
if its from a stop, or above 40,
ill leave it in D
but the real reason why my 98 A4 TA beat the 00 M6 TA WS6,
is because i bought the highest stall i could tolerate
and the stickiest tires i could afford.
if your trans is shifting slowly..
i installed a TransGo shift kit
the high stall absorbed alot of the "kick"
so it doesnt shift as hard as it used to,
but it still seems to shift faster.
dont forget the transmission cooler

What works best for me, is to set my *roll* at 2000 rpm...in almost any gear, I get a great down shift. However I will say that I have a preference for 20, 40, or 60 mph, if on a roll, but my true preference is from a standing start with warm DR's, so I can take full advantage of 1st gear. God, I love that raped ape howl, out of the hole!

With the 4L60E, and you are going to "drag race" it, leave it in OD. This will prevent the overrun clutch assembly from coming on, especially on the 2-3 shift where you need all the oil going to the 3-4 clutches on a WOT shift, and not to the overrun setup also. Dyno tests have shown this to be the best way of doing this with the least amount of problems. A 2-3 flare at WOT in the "D" position, and the flare is not happening in the OD position is indicative of not enough oil going to the 3-4 clutches. This is only "one" of the good reasons why the 3-4 clutch pack will live longer when you leave it in the OD position when drag racing it.

Also, it has been recommended by my tranny builder to leave it in 1st gear for the burnout, not only for a more controllable burnout, but the tranny actually engages an extra clutch for more holding power when shifter is in 1.
