Stock ls6 zo6 heads and ? cam....
I have a 2000 ls1 camaro. Purchased a set of 243's off a ls6 zo6 vette. What would be the largest cam i could go with without having to fly cut the pistons. I also read in another post that the valves are longer from 241's to 243's so is that going to cause me to use a different length pushrod? Im looking for 420rwhp with ls6 intake and longtubes. Any info is helpful.

To answer YOUR question, T-REX from Thunder, but as JPH mentionned this is not the best cam to put in.
To enjoy that cam, you'll need a heavy duty clutch, a stronger differential with 410 gears (or higher), ported TB.
I agree about his cam choice, but a TR230/224 on a 111 will be even better cam only.
Or a custom grind like:
230/228 .612/.588 110-1 LSA

I already have an ls6 intake, Pacesetter LT's, Custom 3"to 3.5" y dumped, volant CAI, and ported TB and MAF. When dynoed as is made 349rwhp. I want it to be driveable but its not a daily driver so big is good i just don't want to have to worry about piston to valve clearence. So a trex would clear with stock 243 heads?

Are there any caveats to using a reverse split cam? I am in the same boat, I found some 243 castings local, and very cheap. I am now looking for a cam, but I don't want to sacrifice the drive ability of my car. However, I don't want to regret picking a cam that is too small either...
