The powerglide will keep you in boost better no matter how quickly you can shift the T56 it will always fall out of boost for a moment.
The glide should be worth close to 1/2 second faster I'm guessing. You also will drop another 60-80lbs in weight as a T56 is one heavy beast with a clutch setup.

Logs show no drop in boost after I hit 2nd. And...it barely falls in the 1-2 shift. With that being said...the car still is unloading slightly causing the tire spin I believe.
No doubt the auto will be better but I think it is all going to be from the line. The power that the auto will eat is going to cause the car to plane out on the big end...right?

The auto will however be faster, quicker, and get better traction.
The only advantage I can see for me with an auto is traction. It's hard for me to understand how it will work going from a 4spd to a 2spd. Right now I have 3 gears to get me to my 1:1 gear. With the glide it will be only 1...seems like some acceleration is left on the table. What first gear set will I need and what rpm converter and etc. I am seriously uneducated on autos and the parts. I see a 1.80 and a 1.76 first gear I think. Wouldn't a 2.xx be a better choice? How much harder will leavin on a transbrake be on my driveline? Sorry if these are stupid questions but I know nothing. Also what kind of upgrades does the glide need?
As I said, I'm not giving up on the t56 yet, I feel like it can work with some changes to the alignment and tires. Maybe a better boost controller. I am going to try and round up so auto stuff though.

Your car is motivating the hell out of me! I'm building a very similar setup for my s13
LQ9/Built T-56
Garrett GT-4780
Walbro 400 with 8an lines, and 100lb injectors
224 cam
6 Point and other supporting mods
Keep the updates coming, and possibly more video's!?

Swapped to a th400 and have been trying to get it lined out. Seems like if I leave under any power it just blows the tires off. Going to soften the suspension some and try to get it to leave better tonight. I'll update if it does anything worth talking about.

Didn't catch what tuning software you are using, if it is standalone you should have the ability to setup flat shifting. If it is on HPTuners you could add a WOT BOX and cut spark on the clutch switch for a split second allowing a WOT shift, TB never closes so you would only loose a lb or two if any.
