One piece block is the way to go. I've always ran them and they need less grinding if your putting a stroker crank in it. Try to get an 880 block they are very strong.

There actually is an adapter to put a two piece rear main seal crank in a one piece block but that is besides the point and seems kinda pointless.
If you are starting from scratch the number one thing you have to think about is machining and clearancing the block. It can get very expensive quickly. With splaid caps and a race prepped block I spent nearly 1100 bucks on my block with hardware.
However GMPP offers a one piece rear main seal 4bolt 350 880 casting block that is bored .030 and is clearanced for a 3.75 stroke with hbeam rods. Its less than 700 bucks ready to wash and assemble.
